The present study describes the improvement of previously identified fungal strain Aspergillus carbonarius for enhancement of pectinase production by inducing mutations using physical and chemical mutagens. Aiming to increase the potentiality in pectinase production, the parental strain was treated for three times with four mutagens UV irradiation, Colchicine, Hydrogen peroxide and Ethidium bromide to obtain mutants. Mutants were selected based on higher enzyme activity, improved growth rates and varied morphology with increased pectinase production. All the surviving mutants were assessed quantitatively after first mutagenic treatment. The stability of the best mutants was tested by repeating the exposures for two times to obtain 3 generation mutants. These mutants were tested quantitatively to assess the pectinase production. Of all the best mutants E8 showed maximum activity producing 65U/ml pectinase enzyme compared to wild and sister mutants. The wild strain of A. carbonarius is a low pectinase producing organism as per literature. This strain was successfully mutated to increase the productivity rate to 1.8 fold in comparison to wild strain. This overproduction and strain stability may be due to repeated mutagenic treatments.

The present work was carried out with an aim to isolate an Exo-pectinase producing fungal strain, to optimize physical factors of the enzyme activity and to formulate a media for the cost effective production of the enzyme. Screening, isolation and identification of Aspergillus tamarii and Aspergillus carbonarius were done. Various process variables like pH, temperature and incubation time of t...

Overexpression of a C4-dicarboxylate transporter is the key for rerouting citric acid to C4-dicarboxylic acid production in Aspergillus carbonarius

BACKGROUND C4-dicarboxylic acids, including malic acid, fumaric acid and succinic acid, are valuable organic acids that can be produced and secreted by a number of microorganisms. Previous studies on organic acid production by Aspergillus carbonarius, which is capable of producing high amounts of citric acid from varieties carbon sources, have revealed its potential as a fungal cell factory. Ea...

ISOLATION, PRODUCTION AND CHARACTERIZATION OF EXTRACELLULAR PECTINASE FROM Aspergillus niger K3

A pectinase producing fungus was isolated from compost soil. Morphological characteristics, microscopic observations and molecular characterization based on internal transcribed spacer (ITS) sequences led to its identification as Aspergillus niger (GenBank Accession Number: JX556221) and it was designated as Aspergillus niger strain K3. In the present study, the culture condition of the media w...

VeA and LaeA transcriptional factors regulate ochratoxin A biosynthesis in Aspergillus carbonarius.

Ochratoxin A (OTA) is a mycotoxin with nephrotoxic, teratogenic and immunotoxic properties which represents a serious risk for human and animal health. Aspergillus carbonarius is considered the main OTA-producing species in grapes and products such as raisins, wine or juices, although it has also been isolated from coffee, cocoa and cereals. Till now not much information is available about regu...

The loss of the inducible Aspergillus carbonarius MFS transporter MfsA leads to ochratoxin A overproduction.

Ochratoxin A (OTA), a nephrotoxic compound produced by certain Aspergillus and Penicillium species, is one of the most abundant mycotoxins in food commodities. Aspergillus carbonarius is the main source of OTA in wine, grape juice and dried vine fruits. Although many studies have focused on OTA production by A. carbonarius, little is known about the genes related to OTA production and transport...
